چکیده
Abstract Background Patient-derived organoids (PDO) have been proposed as a novel in vitro method of drug screening for different types cancer. However, to date, extrahepatic biliary tract carcinoma (eBTC) PDOs not yet fully established. Methods We collected six samples gallbladder (GBC) and one sample cholangiocarcinoma (eCCA) from seven patients attempt establish eBTC screening. successfully established five GBC eCCA PDOs. Histological staining was used compare structural features between the original tissues cancer Whole exome sequencing (WES) performed analyze genetic profiles Drug screening, including gemcitabine, 5-fluorouracil, cisplatin, paclitaxel, infigratinib, ivosidenib, measured verified by clinical effects certain cases. Results Different exhibited diverse growth rates during culture. Hematoxylin eosin demonstrated that structures most retained adenocarcinoma. Immunohistological periodic acid-schiff revealed marker expression similar specimens. Genetic four specimens, well paired PDOs, were analyzed using whole sequencing. Three high degree similarity when compared except GBC2 PDO, which only had concordance 74% proportion single nucleotide polymorphisms coding sequence. In general, gemcitabine found be efficient treatment, it showed moderate or significant inhibitory impact on growth. confirmed extent three Conclusions Our study series contributed field Additional enhancements should explored improve rate preserve their immune microenvironment.

Interventional radiologists fi rst became involved in the management of malignant obstruction of the biliary tree in the late 1960s, when Kaude et al. introduced nonsurgical biliary drainage. Since then, improved diagnostic imaging techniques, significant developments in interventional radiology, and the results of clinical trials have revolutionized and clearly defi ned the role of percutaneou...

gallbladder and biliary tree is low compared with the many parenchymal hepatic conditions that occur in animals. Diseases of the extrahepatic biliary system are often confused with other intraabdominal disorders because their course and clinical signs are similar. متن کاملTargeting mutant RAS in patient-derived colorectal cancer organoids by combinatorial drug screening
Colorectal cancer (CRC) organoids can be derived from almost all CRC patients and therefore capture the genetic diversity of this disease. We assembled a panel of CRC organoids carrying either wild-type or mutant RAS, as well as normal organoids and tumor organoids with a CRISPR-introduced oncogenic KRAS mutation. Using this panel, we evaluated RAS pathway inhibitors and drug combinations that ...

colloid carcinoma, also called mucinous carcinoma, is classified as a histologic variant of adenocarcinoma. because the colloid carcinoma of the biliary tree is exceedingly rare, the imaging characteristics and the clinical features of colloid carcinoma remain relatively unknown. we report a case of colloid carcinoma of the common bile duct and its accompanied metastatic lymphadenopathies with ...

The patient is a previously healthy 52-year-old woman who presented with dyspepsia for two months. Multiple imaging modalities including ultrasound, computed tomography (CT), and magnetic resonance imaging (MRI) showed diffuse bile duct dilatation with an obstructive lesion of the distal extrahepatic biliary duct (EHD) as well as two masses in the peripancreatic area. The peripancreatic masses ...
